Summary

This reagent is a fish serum-based immunoreagent designed for use with chicken antibodies. It already contains concentrated buffer and non-ionic detergent (tween-20), allowing simple dilution with water.

Note :

WB
After transferring proteins from your gel onto nitrocellu-lose, incubate the membrane with diluted GTX85478 for 1 hour at room temperature with gentle agitation. The blots should then be washed for 15 minutes with PBS prior to addition of primary antibody. It is not necessary to dilute your chicken antibodies or secondary anti-chicken IgY reagents with GTX85478 . Instead, dilute these reagents in PBS with 0.1% (w/v) of a non-ionic detergent, such as triton X-100 or TWEEN-20.

ICC/IF
In a manner similar to that described above for western blots, pre-block your paraformaldehyde-fixed cultured cells or tissue sections with diluted GTX85478 for 1 hour at room temperature. Then, wash off the GTX85478 with PBS prior to addition of chicken antibodies (diluted in PBS with 0.1% non-ionic detergent).

IHC
In a manner similar to that described above for western blots, pre-block your paraformaldehyde-fixed cultured cells or tissue sections with diluted GTX85478 for 1 hour at room temperature. Then, wash off the GTX85478 with PBS prior to addition of chicken antibodies (diluted in PBS with 0.1% non-ionic detergent).
Not tested in other applications.

Store at room (ambient) temperature. Do not put in the refrigerator or freezer, otherwise crystals will form which can cause background problems.
