|Application Note||Recommended Starting Dilutions:|
For FACS: Use at a concentration of 5-10 μg per 1 x 106 cells.
For flow cytometry, cells should be fixed in 3.7% formaldehyde for 10 min.at room temperature to permeabilize the cell membrane. It is recommended touse 5-10 ug antibody/one million cells, and incubate for 30-60 minutes at 4degrees C. Please pay attention that the exact antibody concentration andsubsequent steps should be empirically optimized for the system understudy.
For IHC-Fr: Use at a concentration of 5-10 μg/ml.
For IHC-P: Use at a concentration of 1-10 μg/ml.
For WB: Use at a concentration of 1-10 μg/ml. Boiling samples may reduce visibility of the protein.
Optimal dilutions/concentrations should be determined by the researcher.

|Background||P Glycoprotein, the product of the MDR1 gene, is expressed in distinct non-malignant cells, typically cells with secretory and excretory functions. It is assumed to function as an ATP-dependent drug efflux pump with broad substrate specificity. The highest expression of P Glycoprotein has been observed in kidney (proximal tubules), liver (bile canaliculi), adrenal gland and intestine, suggesting that the primary role of P Glycoprotein is in the normal secretion of physiological metabolites and ingested chemicals into bile, urine and the lumen of the intestinal tract. Elevated levels of P Glycoprotein have also been reported in multidrug-resistant cell lines and in colon, endometrial, ovarian, and breast tumors, as well as in sarcomas and leukemias / lymphomas.|

Specific reactivity to human, mouse, rat, dog and primate. Detects bands of ~170kDa (MDR1 P-glycoprotein) and of ~140kDa (MDR3 P-glycoprotein) by Western blot.|
