Methylation of cytosine in genomic DNA plays an important role in the epigenetic modulation of gene expression. Though multiple DNA methyltransferases have been identified and characterized, the identitiy and function of DNA demethylases in mammalian cells have remained elusive. In 2009, tet oncogene 1 (TET1) was identified as a potential candidate effector of DNA demethylation. TET1 is believed to mediate the oxidative demethylation of 5-methylcytosine by creating a 5-hydroxymethylcytosine (5-hmC) intermediate. TET1 was originally identified as a translocation associated with acute myeloid leukemia. Mutations to TET1 have been associated with various malignancies and developmental disorders. TET1 has also recently been implicated in the maintenance of pluripotency in embryonic stem cells due to its role in the demethylation of promoters for pluripotency-associated genes.
