Application Note:
IHC-P (both antibodies, although GTX20199 (NB 100-122) has been used with varying success): Use at a dilution of 1/100 - 1/1000. WB (both antibodies): Use at a dilution of 1/500 - 1/1000. Detects a band of approximately 118 kDa. Nuclear extracts should be used for Western analysis, if possibleNot tested in other applications. Optimal dilutions/concentrations should be determined by the end user.
Background:
Hypoxia contributes significantly to the pathophysiology of major categories of human disease, including myocardial and cerebral ischemia, cancer, pulmonary hypertension, congenital heart disease and chronic obstructive pulmonary disease. HIF2 alpha is predominantly expressed in highly vascularized tissues of adult humans and endothelial cells of the embryonic and adult mouse, whereas HIF1 alpha functions primarily in extravascular tissues. HIF2 alpha is also a potent activator of the tie2 gene, which is known to be selectively expressed in endothelial cells.
Cross Reactivity:
GTX20199 Cross-reacts with Human, Mouse and Rat. Not yet tested in other species. GTX28365 Cross-reacts with Human and Rat. Results in mouse have been mixed (some + and some -). Not yet tested in other species.
Storage Instruction:
Store at 4 °C. Aliquot and store at -20 °C long-term.
