10X Automation Buffer


Background:

The 10X Automation Buffer is a concentrated formulation of a rinsing buffer for use in autostainers employing capillary action technology, as well as for manual immunostaining. It was developed in collaboration with David Brigati M.D., Department of Pathology, University of Oklahoma, Oklahoma City, OK. This buffer is recommended for all the rinsing steps performed when doing immunocytochemical procedures. The 10X Automation Buffer is recommended for all procedures utilizing Dr. David Brigati's capillary action technique for transporting reagents to the specimens. No pH adjustment or additives are required when using this formulation. Simply dilute the concentrated 10-fold buffer with distilled or deionized water. A total of 10 liters of working rinsing solution can be prepared with 1 liter of the concentrated reagent. This buffer is also the reagent of choice for rinsing tissue sections that are subjected to immunocytochemical staining using conventional horizontal processing techniques. This concentrated buffer is recommended for the rinsing steps when performing most immunoassays including immunohistochemical staining of tissue sections using any of our HistoScan(tm), LymphoScan(tm) or BioStain(tm) Super ABC kits. Seven Step Procedure: 1.After the immunostaining, rinse slides with distilled water and drain. 2.Introduce the hematoxylin. 3.Incubate the slides for 1-2 min. at room temperature. 4.Drain off the hematoxylin and wash slides several times with distilled water. 5.Incubate the slides with 1X Automation Buffer or PBS (pH7.4) for 1 minute. 6.Wash with distilled water. 7.Mount with Crystal/Mount (Cat. #M02 or #M03). TBS based (Tris-Buffered Saline): -- Deionized or Distilled Water -- 50mM Tris Base -- 150mM Sodium Chloride (NaCl) -- 0.02% Non-ionic detergent (Triton-X 100, NP-40, Tween-20 or BRIG 35.) detergent used to reduce background and aid in breaking of surface tension for capillary action -- Preservative --The pH of 10X buffer is 7.5, when diluted 10 times, the pH of working solution will be 7.5+/- 0.1 Alternatively: -- Substitute 10mM Phosphate for Tris Base. -- Substitute 10mM Phosphate for Tris Base.

Storage Instruction:

Store at room temperature.

Tested Applications:

Immunohistochemistry. The usefulness of this product in other applications has not been determined.
