Auto / Iodine


Background:

Tissues subjected to mercury containing fixatives, such as Zenker and B5, require treatment with iodine solution in order to remove the mercuric pigments. Traditionally, following iodine treatment, sections are treated with a diluted solution of sodium thiosulfate. New Auto/Iodine, developed in collaboration with David Brigati, M.D., eliminates the need to use sodium thiosulfate treatment, as well as initiates the process of rehydration. It removes mercuric salts in a single step procedure. Tissues subjected to fixatives containing mercury salts, such as Zenker and B5, require treatment with an iodine solution in order to remove the salts introduced during fixation. Typically, following the iodine treatment, the sections are treated with a diluted solution of sodium thiosulfate. Auto/Iodine is a new formulation developed for automated immunocytochemistry by David Brigati, M.D. This formulation eliminates the need to use the thiosulfate treatment, as it removes mercuric salts in a single step procedure. Auto/Iodine is used following the last alcohol step in the deparaffinization process and it does not cause the detachment of mounted tissue specimens. The reagent is supplied in a 500 mL size and is very stable when stored at room temperature. Step-by-step Procedure: 1.Deparaffinize the sections. 2.Bring the sections to 100% alcohol. 3.Blot 100% alcohol and introduce Endo/Blocker (Cat. No. GTX73347). 4.Incubate for 5 minutes at room temperature. 5.Blot Endo/Blocker and introduce Auto/Iodine solution. 6.Incubate for one minute at room temperature and blot. 7.Introduce 10X Automation Buffer (Cat. No. GTX73344). Incubate for one minute and blot. 8.Repeat step 7 once more and proceed with the staining protocol. MicroProbe Prodedure: Replace the 70% alcohol in station 3 with the Auto/Iodine solution and follow the protocol mentioned above. After Xylene, bring the sections to 100% alcohol and then incubate in Endo/Blocker to neutralize endogenous peroxidase. Next, treat the slides with Auto/Iodine and then go directly to 10X Automation Buffer. Code-On Automated Procedure: Dedicate a reagent bucket station to the Auto/Iodine solution. Introduce the above mentioned protocol into the software that you are already using. Appearance:: Precipitation may occur in this reagent after prolonged storage. This should not interfere with the function of this product. If so desired, it can be filtered on filter paper in a well ventilated hood.
