Proximity ligation assay (PLA) is a sensitive and specific tool to detect endogenous proteins, protein-protein interactions, and protein posttranslational modifications at single-molecule resolution. PLA is based on dual recognition of the target protein by a pair of oligonucleotide-conjugated antibodies. With the addition of connecting oligos, the two oligonucleotides form a DNA circle that subsequently serves as a template for localized rolling circle amplification (RCA) (see figure above). An otherwise weak signal from a low-abundance protein or a rare event such as a particular protein-protein interaction or protein modification can be amplified over 1,000-fold in situ by RCA, thus making PLA a valuable research and diagnostic tool. PLA can be performed on adherent cell or tissue section samples.
