JNJ-A07, a pan-serotype dengue virus (DENV) inhibitor, has demonstrated therapeutic activity against DENV infection in murine models. Though it is known to block the interaction between NS2B-NS3 and NS4B-containing proteins, its exact mechanism of action had not been discerned. Now, Kiemel et al. report that JNJ-A07 targets the interaction between NS2B-NS3 and the NS4A-2K-NS4B precursor, thereby preventing de novo formation of vesicle packet structures that are indispensable for viral RNA replication. In addition to providing a better understanding of JNJ-A07 activity, this work may also offer insight into the mechanisms-of-action of other NS4B-directed agents.
