As is commonly seen with membrane proteins, significant hydrophobicity can lead to aggregation following boiling of samples prior to SDS-PAGE and subsequent western blotting. We recommend to avoid boiling in this case.

Background

This gene encodes a G-protein coupled receptor that binds neuropeptides of the corticotropin releasing hormone family that are major regulators of the hypothalamic-pituitary-adrenal pathway. The encoded protein is essential for the activation of signal transduction pathways that regulate diverse physiological processes including stress, reproduction, immune response and obesity. Alternative splicing results in multiple transcript variants.

U87-MG whole cell and membrane extracts (30 μg) were separated by 10% SDS-PAGE, and the membrane was blotted with CRHR1 antibody (GTX130244) diluted at 1:1000. The HRP-conjugated anti-rabbit IgG antibody (GTX213110-01) was used to detect the primary antibody.
