Tropomyosin antibody [CH1]

Cat No. GTX27786

Host Mouse
Clonality Monoclonal
Clone Name CH1
Isotype IgG1
Application WB, IHC-P, IHC-Fr, IP, RIA
Reactivity Human, Rat
APPLICATION

Application Note

This antibody can be used on paraffin-embedded, formalin-fixed human heart and tongue tissuesections. Prolonged fixation in buffered formalin can destroy the epitope. Other applications for this antibody include radioimmunoassays, immunoblotting (1:50-1:100)and immunoprecipitation.

Positive Control

Human tongue, heart and chicken heart

Specificity/Sensitivity

This antibody is specific for human striated muscle forms of tropomyosin, including cardiac alpha tropomyosin and skeletal muscle forms of tropomyosin. This rod-shaped protein is associated with contractile tissue, where it regulates the Ca 2+ -dependent interaction between actin and myosin. This antibody reacts with human striated muscle forms of tropomyosin. It also cross-reacts with chicken cardiac muscle. This antibody does not cross-react with smooth muscle or non-muscle isoforms of tropomyosin.

TARGET

Background

Tropomyosin is a dimeric coiled-coil protein that binds along the length of actin filaments. It is associated with the thin filaments of muscle cells and the microfilaments of nonmuscle cells. Chicken embryo fibroblasts (CEF) contain five isoforms of tropomyosin (a, b, 1, 2, and 3), identified as such by their different apparent molecular masses after separation by SDS-PAGE, but similar biochemical properties, such as resistance to heat and organic solvents, the ability to bind to F-actin filaments, and the lack of proline and tryptophan.
