APPLICATION

Application Note

This antibody can be used on formalin-fixed, paraffin-embedded tissue sections at a dilution of 1:50-1:75 . Heat mediated antigen retrieval is strongly recommended for consistent and reproducible results. For western blotting use at 1:100-1:250.This antibody is not suitable for frozen tissue sections. The optimal conditions should be determined by the individual laboratory.

Breast Tumor - 50% of breat tumors are p53 positive, especially those lacking estrogen and/or progesterone receptors and with high proliferation index.

Product Note

This antibody reacts with wild type as well as mutant type of the p53 protein. It recognizes an epitope in the N-terminus of the human p53 protein. This antibody recognizes cell nuclei in which the p53 protein are generally localized. This antibody does not react with normal fibroblasts.

Background

This gene encodes a tumor suppressor protein containing transcriptional activation, DNA binding, and oligomerization domains. The encoded protein responds to diverse cellular stresses to regulate expression of target genes, thereby inducing cell cycle arrest, apoptosis, senescence, DNA repair, or changes in metabolism. Mutations in this gene are associated with a variety of human cancers, including hereditary cancers such as Li-Fraumeni syndrome. Alternative splicing of this gene and the use of alternate promoters result in multiple transcript variants and isoforms. Additional isoforms have also been shown to result from the use of alternate translation initiation codons from identical transcript variants (PMIDs: 12032546, 20937277).
