As is commonly seen with membrane proteins, significant hydrophobicity can lead to aggregation following boiling of samples prior to SDS-PAGE and subsequent western blotting. We recommend to avoid boiling in this case.

Background

This gene encodes a transmembrane protein that plays a key regulatory role in the process of autophagy. The ectopic overexpression of the encoded protein in cultured cells triggers autophagy even under nutrient-rich conditions. This gene is overexpressed in pancreatitis affected acinar cells where the encoded protein mediates sequestration and degradation of potentially deleterious activated zymogen granules in a process termed, zymophagy. Unboiled HepG2 whole cell and membrane extracts (30 μg) were separated by 10% SDS-PAGE, and the membrane was blotted with TMEM49 antibody (GTX131510) diluted at 1:1000. The HRP-conjugated anti-rabbit IgG antibody (GTX213110-01) was used to detect the primary antibody. (WCE: whole cell extract; ME: membrane extract)
