| Product Name | Antigen retrieval solution |
| Product Description | Antigen retrieval solution-enzymatic (pepsin) |
| Background | Most fixatives immobilize the tissue antigens by cross linkages. Sometimes, extensive cross linking of proteins in high concentrations can also lead to the masking or destruction of antigens, which leads to poor staining or no staining at all. This loss of staining can be restored to a certain extent by pretreatment of tissue sections with proteolytic enzymes. The unmasking of tissue sections is an important requirement for the optimal staining of antigens like immunoglobulins, keratins, carcinoembryonic antigen, factor VIII related antigen, testosterone, and to a lesser extent S100 protein. |
| Applications | IHC (Formalin-fixed paraffin-embedded sections). The usefulness of this product in other applications has not been determined. |
| Application Note | This antigen retrieval solution is provided in a ready-to-use format requiring no further preparation. 1.) Apply this solution to tissue sections following their initial hydration step. Incubate for 10 min at 37°C or 45°C (this incubation time may vary depending on the degree of fixation of the tissue sections. 2.) Wash tissue sections well with IHC wash buffer (GTX28195), or PBS or TBS if immunostaining by hand. |
| Form Supplied | Liquid |
| Storage Instruction | Refridgerate at 4°C. Do not freeze. Avoid extended exposure to room temperature. |
| Notes | This Antigen retrieval solution is a stabilised enzymed designed for the uncovering of tissue antigens fixed with formalin. Do not use this antigen retrieval solution when detecting Cytokeratins using the AE3 monoclonal (GTX20870, GTX29287, GTX20961): instead use Antigen Retrieval Kit enzymatic (GTX28212).This product was previously known as Pepsin reagent for unmasking." |
