Summary

Proteinase K is specifically formulated to achieve proteolytic digestion of formalin fixed, paraffin embedded tissue sections prior to performing in situ hybridization procedures. Proteolytic digestion of formalin fixed tissues improves the accessibility of target DNA to biotinylated probes, which results in better in situ staining.

APPLICATION

Application Note

Preparation of working solutions :
1. Place 100μl Proteinase K solution in a glass tube, and 2.4ml distilled water and mix.
Protocol Recommendations :
1. Optimal digestion conditions will vary depending on fixation conditions and should be optimized by the end user.
2. Incubation of tissue sections for 10 - 30 minutes at 37ºC is sufficient for in situ.

Storage

Store at 2-8ºC. Proteinase K is stable for 6 months. For long term storage, aliqout Proteinase K into smaller sizes and store at -20ºC. Do not use after expiration date printed on label.
