Background

Trident plus Western HRP Substrate is a luminol-based, chemiluminescent substrate for the detection of HRP-conjugated antibody on immunoblot at picogram level. Trident plus Western HRP Substrate produces an excellent signal that can be easily detected by chemiluminescent imaging methods including X-ray film and chemiluminescence imaging system.
User instruction: use 1 to 1 ratio of the mixture of solution A and B. Keep the mixture on ice and immediately apply to the blot. Shortly after the immersion of the blot with the mixed solution, the blot would be ready to proceed to the film or CCD imaging.

Recombinant protein mcherry is loaded as the follows:
Lane A : 50 ng recombinant protein
Lane B : 25 ng recombinant protein
Lane C : 12.5 ng recombinant protein
Lane D : 6.25 ng recombinant protein
Lane E : 3.13 ng recombinant protein
Lane F : 1560 pg recombinant protein
Lane G : 780 pg recombinant protein
Lane H : 390 pg recombinant protein
Lane I : 195 pg recombinant protein
Lane J : 97.5 pg recombinant protein
The blot was probed with mcherry antibody (GTX128508) diluted at 1:2000. A HRP-conjugated secondary antibody was applied and developed with Trident plus Western HRP Substrate (GTX400006) and regular ECL. Exposure time : 18 seconds, 90 seconds, and 180 seconds.
