Manual staining : Heat-induced antigen retrieval for 5 minutes in an autoclave at 121ºC in pH 7.8 Tris-EDTA-based Target Retrieval Solution buffer. Based on the testing result, the pH 9.0 is optimal but pH 7.8 results in only slightly weaker stainings. At pH 6.0, the immunostaining is markedly reduced.
This antibody has been validated by Autostainer including Agilent/Dako - Autostainer Link 48 and BOND RX - Leica. The detailed steps by Autostainer please refer to the protocol.

Positive Control
Tonsil or appendix: A strong, predominantly membranous staining of the mantle zone B-cells, the germinal centre B- cells and the interfollicular B-cells is required, while all other cells should stain negative.
Negative Control
Tonsil or appendix: A large fraction of the interfollicular lymphocytes and all epithelial cells must not show any staining.

Background
This gene encodes a member of the membrane-spanning 4A gene family. Members of this nascent protein family are characterized by common structural features and similar intron/exon splice boundaries and display unique expression patterns among hematopoietic cells and nonlymphoid tissues. This gene encodes a B-lymphocyte surface molecule which plays a role in the development and differentiation of B-cells into plasma cells. This family member is localized to 11q12, among a cluster of family members. Alternative splicing of this gene results in two transcript variants which encode the same protein.
