As is commonly seen with membrane proteins, significant hydrophobicity can lead to aggregation following boiling of samples prior to SDS-PAGE and subsequent western blotting. We recommend to avoid boiling in this case.

Background

Fractalkine is a transmembrane protein and chemokine involved in the adhesion and migration of leukocytes. The protein encoded by this gene is a receptor for fractalkine. The encoded protein also is a coreceptor for HIV-1, and some variations in this gene lead to increased susceptibility to HIV-1 infection and rapid progression to AIDS. Four transcript variants encoding two different isoforms have been found for this gene. [provided by RefSeq]

Non-transfected (–) and transfected (+) boiled and unboiled 293T whole cell extracts (30 μg) were separated by 10% SDS-PAGE, and the membrane was blotted with CX3CR1 antibody (GTX130027) diluted at 1:1000. The HRP-conjugated anti-rabbit IgG antibody (GTX213110-01) was used to detect the primary antibody.

CX3CR1 antibody detects CX3CR1 protein at cell membrane and cytoplasm by immunofluorescent analysis.
Sample: THP-1 cells were fixed in 4% paraformaldehyde at RT for 15 min.
Green: CX3CR1 stained by CX3CR1 antibody (GTX130027) diluted at 1:500.
Blue: Fluoroshield with DAPI (GTX30920).
