Background

This gene encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This mitochondrial protein initiates the degradation of 1,25-dihydroxyvitamin D3, the physiologically active form of vitamin D3, by hydroxylation of the side chain. In regulating the level of vitamin D3, this enzyme plays a role in calcium homeostasis and the vitamin D endocrine system. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

KO/KD

Non-transfected (–) and transfected (+) A549 whole cell extracts (30 μg) were separated by 7.5% SDS-PAGE, and the membrane was blotted with CYP24A1 antibody [HL1783] (GTX637435) diluted at 1:1000. The HRP-conjugated anti-rabbit IgG antibody (GTX213110-01) was used to detect the primary antibody, and the signal was developed with Trident ECL plus-Enhanced.

Various whole cell extracts (30 μg) were separated by 10% SDS-PAGE, and the membrane was blotted with CYP24A1 antibody [HL1783] (GTX637435) diluted at 1:1000. The HRP-conjugated anti-rabbit IgG antibody (GTX213110-01) was used to detect the primary antibody.

Sample: A549 cells were fixed in 4% paraformaldehyde at RT for 15 min.
Green: CYP24A1 stained by CYP24A1 antibody [HL1783] (GTX637435) diluted at 1:500.
Red: alpha Tubulin, a cytoskeleton marker, stained by alpha Tubulin antibody [GT114] (GTX628802) diluted at 1:1000.
Blue: Fluoroshield with DAPI (GTX30920).
