Western Blotting Analysis: 0.5 μg/mL from a representative lot detected Cas9 in 1 μg of Myc-Cas9 expressing HEK293 cell lysate. Immunoprecipitation Analysis: A representative lot immunoprecipitated Cas9 in Flag-Cas9 and Myc-Cas9 expressing cell lysate (performed by an independent laboratory).

IP analysis was performed with lysate from HEK293T cells expressing Flag-tagged S. pyogenes Cas9 and either Cas9 antibody [7A9] followed by incubation with a 1:1 mixture of protein A/G sepharose beads (Cas9 Sup) or Cas9 antibody [7A9] crosslinked with a 1:1 mixture of protein A/G sepharose beads (Cas9 X-link). The precipitates were detected by the same antibody. Asterisk indicates IgG heavy chain.
