APPLICATION

Application Note

*Optimal dilutions/concentrations should be determined by the researcher.
Application Dilution
WB Assay dependent
ICC/IF 1:250
FACS 1:20
IHC Assay dependent
Not tested in other applications.

Calculated MW

134 kDa. ( Note )

Specificity/Sensitivity

The full-length EGFR includes the 24 aa signal peptide that is cleaved during EGFR maturation. The Tyr1148 of mature EGFR is equivalent to Tyr1172 of uncleaved EGFR.
PROPERTIES

Form

Liquid

Buffer

PBS pH 7.3, 1mg/ml BSA, 50% glycerol, 0.05% sodium azide

Storage

Store as concentrated solution. Centrifuge briefly prior to opening vial. For short-term storage (1-2 weeks), store at 4ºC. For long-term storage, aliquot and store at -20ºC or below. Avoid multiple freeze-thaw cycles.

Antigen Species

Human

Immunogen

The antiserum was produced against a chemically synthesized phosphopeptide derived from the region of EGFR that contains tyrosine 1148. The sequence is conserved in mouse, rat and chicken.

Purification

Purified by antigen-affinity chromatography

Conjugation

Unconjugated

Background

The protein encoded by this gene is a transmembrane glycoprotein that is a member of the protein kinase superfamily. This protein is a receptor for members of the epidermal growth factor family. EGFR is a cell surface protein that binds to epidermal growth factor. Binding of the protein to a ligand induces receptor dimerization and tyrosine autophosphorylation and leads to cell proliferation. Mutations in this gene are associated with lung cancer. Orthogonal

WB (peptide competition) analysis of A431 cells stimulated with 200 ng/mL EGF (Lane 2-5) for 15 minutes using GTX10643 EGFR (phospho Tyr1148) antibody prior incubated with, non-phosphopeptide corresponding to the phosphopeptide immunogen (Lane 3), a generic phosphotyrosine-containing peptide (Lane 4), or the phosphopeptide immunogen (Lane 5) control. The data show that only the immunogen phosphopeptide blocks the signal, demonstrating the specificity of the antibody.
