Application Note

Recommended Starting Dilutions:
For WB: Use at a dilution of 1:100-1:3000. Recognizes a band at 118kDa.
For IHC: Use at a dilution of 1:100.
For ICC/IF: Use at a dilution of 1:100-1:1000.

Background

Hypoxia contributes significantly to the pathophysiology of major categories of human disease, including myocardial and cerebral ischemia, cancer, pulmonary hypertension, congenital heart disease and chronic obstructive pulmonary disease. HIF-2 alpha is predominantly expressed in highly vascularized tissues of adult humans and endothelial cells of the embryonic and adult mouse, whereas HIF-1alpha functions primarily in extravascular tissues. HIF-2 alpha is also a potent activator of the tie-2 gene, which is known to be selectively expressed in endothelial cells. (1)

HIF2 alpha antibody detects HIF2 alpha protein by western blot analysis. Un-treated (-) and treated (+, 0.01 O2 treatment for 24h) HepG2 whole cell extracts (30 µg) were separated by 7.5% SDS-PAGE, and the membrane was blotted with HIF2 alpha antibody (GTX30114) diluted by 1:500. The HRP-conjugated anti-rabbit IgG antibody (GTX213110-01) was used to detect the primary antibody.

Overexpression

HIF2 alpha antibody detects HIF2 alpha protein by western blot analysis. Non-transfected (-) and HIF2A-transfected (+, including Myc-DDK-tag) 293T whole cell extracts (30 µg) were separated by 5% SDS-PAGE, and the membrane was blotted with HIF2 alpha antibody (GTX30114) diluted by 1:1000. The HRP-conjugated anti-rabbit IgG antibody (GTX213110-01) was used to detect the primary antibody.
