ICC/IF analysis of 4% paraformaldehyde fixed HeLa cells using GTX60812 Histone H2A.Z antibody - ChIP grade. Staining of the cells with the H2A.Z antibody after incubation of the antibody with 10 ng/μl of the immunogen peptide (figure 5B) and with a peptide containing a sequence from the central part of the H2A.Z protein (figure 5C).

ChIP analysis of sheared chromatin from 1x10⁵ K562 cells using GTX60812 Histone H2A.Z antibody - ChIP grade. The IP'd DNA was subsequently analysed with an Illumina Genome Analyzer. Library preparation, cluster generation and sequencing were performed according to the manufacturer's instructions. The 36 bp tags were aligned to the human genome (hg19) using the ELAND algorithm. Figure 2 shows the peak distribution in four genomic regions including the regions surrounding the EIF2S3 and c-fos positive control genes on chromosome X and 14, respectively (figure 2A and B).

ChIP analysis of sheared chromatin from 10⁶ HeLa cells using GTX60812 Histone H2A.Z antibody - ChIP grade. A titration of the antibody consisting of 0.2, 0.5, 1 and 2 μg per ChIP experiment was analysed. IgG (1 μg/IP) was used as negative IP control. Quantitative PCR was performed with primers specific for the promoter of the active genes c-fos and EIF2S3, used as positive controls, and for the coding region of the inactive MB gene and the Sat2 satellite repeat, used as negative controls. This figure shows the recovery, expressed as a % of input (the relative amount of immunoprecipitated DNA compared to input DNA after qPCR analysis).
