Background
This gene encodes a nuclear-localized E3 ubiquitin ligase. The encoded protein can promote tumor formation by targeting tumor suppressor proteins, such as p53, for proteasomal degradation. This gene is itself transcriptionally-regulated by p53. Overexpression or amplification of this locus is detected in a variety of different cancers. There is a pseudogene for this gene on chromosome 2. Alternative splicing results in a multitude of transcript variants, many of which may be expressed only in tumor cells. Untreated (–) and treated (+) HepG2 whole cell extracts (30 μg) were separated by 7.5% SDS-PAGE, and the membrane was blotted with MDM2 antibody (GTX70278) diluted at 1:1000. The HRP-conjugated anti-mouse IgG antibody (GTX213111-01) was used to detect the primary antibody, and the signal was developed with Trident ECL plus-Enhanced.
Multiple bands were observed 60 ~ 97 kDa. It is possibly due to alternative splicing. Reference:PMID: 9840926
