0.05% Sodium azide

Storage

Store as concentrated solution. Centrifuge briefly prior to opening vial. For short-term storage (1-2 weeks), store at 4ºC. For long-term storage, aliquot and store at -20ºC or below. Avoid multiple freeze-thaw cycles.

Concentration

Batch dependent (Please refer to the vial label for the specific concentration.)

Antigen Species

Human

Immunogen

The antiserum was produced against a chemically synthesized phosphopeptide derived from a region of human Rb that contains serine 249 and threonine 252 (based on Swiss Protein database, accession number P06400). The sequence is conserved in human, mouse and rat.

Background

The protein encoded by this gene is a negative regulator of the cell cycle and was the first tumor suppressor gene found. The encoded protein also stabilizes constitutive heterochromatin to maintain the overall chromatin structure. The active, hypophosphorylated form of the protein binds transcription factor E2F1. Defects in this gene are a cause of childhood cancer retinoblastoma (RB), bladder cancer, and osteogenic sarcoma. [provided by RefSeq, Jul 2008]

GTX24788 WB Image

WB analysis of whole cell extracts (30 μg lysate) of Jurkat (Lane 1), Serum Starved Jurkat (Lane 2), Serum Starved for overnight following by serum release (Lane 3), and Jurkat treated for 15 min with 25 ng/ml of Calyculin A (lane 4) using GTX24788 Rb (phospho Ser249) antibody.

Anti-Rb (phospho Ser249) antibody used in Western Blot (WB). GTX24788

GTX24788 WB Image

WB (peptide competition) analysis of Jurkat cells in high growth phase using GTX24788 Rb (phospho Ser249) antibody prior incubated with a generic phosphoserine-containing peptide (Lane 2), a generic phosphothreonine- containing peptide (Lane 3), the non-phosphopeptide corresponding to the phosphopeptide immunogen (Lane 4), or the phosphopeptide immunogen (Lane 5). The data show that only the immunogen phosphopeptide blocks the signal, demonstrating the specificity of the antibody.
