Thrombospondin represents about 25% of the protein content of platelet a-granules and is secreted at sites of platelet activation and aggregation suggesting that the protein has an important role in hemostasis. Given the involvement of TSP in the processes of chemotaxis, adhesion, proliferation and differentiation of leukocytes, fibroblasts, smooth muscle and endothelial cells, it is likely that TSP secreted at sites of wounding or clotting plays several important roles in this provisional matrix facilitating the healing process. The mature TSP peptide chain consists of 1152 amino acids and displays an apparent molecular weight of about 170 to 180kD on reducing SDS gels. The trimer of identical subunits of TSP is formed by interchain disulfides.
