Background

The Tet repressor protein (TetR) regulates transcription of a family of tetracycline resistance determinants in Gram-negative bacteria. The Tet system is the most widely used regulatory system for conditional gene expression in eukaryotic cells. TetR can be used for that purpose in some organisms without further modifications. In mammals and in a large variety of other organisms, however, eukaryotic transcriptional activator or repressor domains are fused to TetR to turn it into an efficient regulator.

Non-transfected (–) and transfected (+) 293T whole cell extracts (30 μg) were separated by 12% SDS-PAGE, and the membrane was blotted with Tetracyclin repressor (tetR) antibody (GTX131984) diluted at 1:10000. The HRP-conjugated anti-rabbit IgG antibody (GTX213110-01) was used to detect the primary antibody.

Indirect ELISA analysis performed by coating plate with recombinant Tet Repressor protein (162.21-2.53 nM). Coated protein was probed with Tet Repressor antibody (GTX70489) (1 μg/mL). Goat anti-rabbit IgG antibody (HRP) (GTX213110-01) (1:10000) was used to detect bound primary antibody.
