Application Note

For WB: Use at a dilution of 1:500-1:10000, For IP: Use at a dilution of 1:100-1:500 , For IHC-P: Use at a dilution of 1:1,000. Antigen retrieval can be done with EDTA at pH8.0 - Some customers have stated that the product works well without antigen retrieval. TARGET

Background

The p53 antibody, clone DO1, specifically recognizes an epitope at the N-terminal end of the human p53 protein between amino acids 20-25. p53 is a tumor suppressor gene that is mutated in a wide variety of sporadic and familial cancers. And p53, as a nuclear protein, mediates cell cycle arrest in response to DNA damage and therefore is important in the maintenance of genomic stability. Recent results indicate an enhanced phosphorylation of p53 by ATM in response to DNA damage on SER15, a mutation which reduces the ability of p53 to arrest cell growth.
