Ubiquitination is catalyzed by E1, E2, and E3 enzymes that, respectively, activate, conjugate, and ligate ubiquitin to substrate proteins. UBE3A, also known as E6AP (E6-associated protein), is a multifunctional E3 ubiquitin ligase (1). Aberrant regulation of UBE3A is involved in oncogenesis, neurodevelopmental disorders, and other human diseases. In addition to its ubiquitin ligase activity, UBE3A also functions as a transcriptional coactivator of steroid hormone receptors. Its direct link to Angelman syndrome and other pathological conditions makes UBE3A a focus of intense basic and clinical research.
