Application Note

For ELISA: Use at a dilution of 1:5,000-1:20,000. For ICC/IF: Use at a dilution of 1:100-1:1000. For WB: Use at a dilution of 1:1000-1:10000. Optimal dilutions/concentrations should be determined by the researcher.

Rabbits were immunized with xxxDDDDK-peptide conjugated to KLH. After multiple immunizations in Freund's adjuvant, serum was collected and the antibodies were affinity purified using the peptide immobilized on a solid phase.

TARGET

Background

Epitope tags provide a method to localize gene products in a variety of cell types, study the topology of proteins and protein complexes, identify associated proteins, and characterize newly identified, low abundance or poorly immunogenic proteins when protein specific antibodies are not available. Tagging with xxxDDDDK may be done at the N-terminus, N-terminus preceded by a methionine residue, C-terminus, and in internal positions of the target protein. The small size of the epitope tag and its high hydrophilicity tend to decrease the possibility of interference with protein expression, proteolytic maturation, antigenicity and function. The enterokinase cleavage site allows it to be completely removed from the purified fusion proteins.

An ELISA plate is coated with 50 μL of a DDDDK tag conjugated BSA at concentration of 10 μg/mL. The peptide conjugated carrier protein is coated and detected with anti-DDDDK tag antibody (GTX77457) at concentration ranged from 1.95 to 125 ng/mL.

Non-transfected (–) and transfected (+) 293T whole cell extracts (30 μg) were separated by 10% SDS-PAGE, and the membrane was blotted with DDDDK tag antibody (GTX77457) diluted at 1:5000. The HRP-conjugated anti-rabbit IgG antibody (GTX213110-01) was used to detect the primary antibody.
