Manual staining : Heat-induced antigen retrieval for 5 minutes in an autoclave at 121ºC in pH 7.8 Tris-EDTA-based Target Retrieval Solution buffer.
This antibody has been validated by Autostainer including Agilent/Dako - Autostainer Link 48 and BOND RX - Leica. The detailed steps by Autostainer please refer to the protocol.

Positive Control
Kidney: A moderate to strong nuclear staining reaction should be seen in a fraction of collecting duct cells and podocytes in glomeruli. ; Tonsil: The vast majority of T helper cells (Th2) in the T-zones should show a weak to moderate nuclear staining.
Negative Control
Kidney: Staining should be absent in proximal and distal tubuli as well as in blood vessels. ; Tonsil: Staining should be absent in B-cells.

Background
This gene encodes a protein which belongs to the GATA family of transcription factors. The protein contains two GATA-type zinc fingers and is an important regulator of T-cell development and plays an important role in endothelial cell biology. Defects in this gene are the cause of hypoparathyroidism with sensorineural deafness and renal dysplasia. [provided by RefSeq, Nov 2009]
