Manual staining : Heat-induced antigen retrieval for 5 minutes in an autoclave at 121ºC in pH 9.0 Tris-EDTA-based Target Retrieval Solution buffer. Based on the testing result, the pH 9.0 is optimal but pH 7.8 results in only slightly weaker stainings. At pH 6, the immunostaining is markedly reduced.
This antibody has been validated by Autostainer including Agilent/Dako - Autostainer Link 48, Bond RX-Lecia, and Roche-Ventana Discovery ULTRA. The detailed steps by Autostainer please refer to the protocol.

Positive Control
Tonsil: A fraction of squamous epithelial cells of the tonsil crypts should show strong mesothelin immunostaining. Fallopian tube: An at least moderate mesothelin positivity should be seen at the apical membrane of epithelial cells.
Negative Control
Tonsil: Squamous epithelial cells of the tonsil surface and all lymphocytes must not show mesothelin immunostaining.

Background
This gene encodes a preproprotein that is proteolytically processed to generate two protein products, megakaryocyte potentiating factor and mesothelin. Megakaryocyte potentiating factor functions as a cytokine that can stimulate colony formation of bone marrow megakaryocytes. Mesothelin is a glycosylphosphatidylinositol-anchored cell-surface protein that may function as a cell adhesion protein. This protein is overexpressed in epithelial mesotheliomas, ovarian cancers and in specific squamous cell carcinomas. Alternative splicing results in multiple transcript variants, at least one of which encodes an isoform that is proteolytically processed. [provided by RefSeq, Feb 2016]
