Manual staining : Heat-induced antigen retrieval for 5 minutes in an autoclave at 121ºC in pH 9.0 Tris-EDTA-based Target Retrieval Solution buffer. Based on the testing result, the pH 9.0 showed a strongest staining pattern. The pH 7.8 is acceptable but lower pH results in a significant reduction of sensitivity.
Not tested in other applications.

Positive Control

Tonsil: All cells of the surface squamous epithelium and most squamous cells of the crypts should show strong KRT15 staining.

Negative Control

Tonsil: All lymphocytes and blood vessels must not show any KRT15 staining.

Background

The protein encoded by this gene is a member of the keratin gene family. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. Most of the type I cytokeratins consist of acidic proteins which are arranged in pairs of heterotypic keratin chains and are clustered in a region on chromosome 17q21.2.
