Glutamic acid decarboxylase (GAD) catalyzes the decarboxylation of glutamate to generate gamma-aminobutyric acid (GABA), the major inhibitory neurotransmitter controlling neuronal excitability. In humans, GAD is expressed by two independently regulated genes (GAD1 and GAD2) to produce two isoforms: GAD67 (67 kDa) and GAD65 (65 kDa), respectively. GAD67 and GAD65 share high protein homology, and both are targeted to mark GABAergic neurons. However, though they both perform the same enzymatic reaction, they have different functional roles and produce GABA in distinct cellular locations (1).
