Background

This gene encodes a member of the aldo/keto reductase superfamily, which consists of more than 40 known enzymes and proteins. These enzymes catalyze the conversion of aldehydes and ketones to their corresponding alcohols by utilizing NADH and/or NADPH as cofactors. The enzymes display overlapping but distinct substrate specificity. This enzyme catalyzes the reaction of progesterone to the inactive form 20-alpha-hydroxy-progesterone. This gene shares high sequence identity with three other gene members and is clustered with those three genes at chromosome 10p15-p14. [provided by RefSeq]

Primary Antibody : 1:5000, diluted in IBB buffer, 25°C, 1Hr
Figure Note : 5μL of Dynabeads protein G and 10μg of AKR1C1 antibody (GTX105620) were added to 200μL of PBS containing 0.02% tween-20, and were mixed by inverting for 1 hr. The pellet of HepG2 cell lysate was crosslinked with the antibody-dynabeads complex by mixing. After the pellet was washed with IP buffer containing PBS including 3% triethanolamine, it was diluted in 20% (w/v) SDS as IP samples.IP sample was eluted in 30μl of the buffer and then 20μl was loaded on the SDS-PAGE. That equals to a two-thirds of 500μg original lysate. For the HepG2 input lysate, the loaded amount was 10μg.
