Note : WB SDS-PAGE under non-denatured condition. Block blot with 1% skimmed milk for 1h and incubate with RM3/1 in 0.1%BSA/TBS for 1h. Detection of ~130 kDa product in spleens extracted in presence of NP-40 or in glucocorticoid-treated human monocytes. ICC/IF Cells were plated on coverslips and fixed in 4% paraformaldehyde for 2 mins. FACS Permeabilization as well as no permeabilization is used. Block cells with 1% BSA for 30mins at 4ºC IP 10 μg RM3/1 was covalently coupled to beads (with 20 mM dimethyl pimelimidate), blocked with 0.2 M glycine buffer for 2 hr and incubated with cell lysate for 2 hr. Neutralizing/Inhibition Reduction of CD69 expression by adding 2.5 μg/ml RM3/1 to 5x10⁵ human T-lymfocytes in presence of exogenous CD163.

Not tested in other applications.

The RM3/1 antigen expression is restricted to human monocytes and macrophages that evolve in the late phase of inflammation, predominantly macrophages of healing tissue.
We do not recommend use of this product for Rat,Guinea pig,Pig samples.

Background

The protein encoded by this gene is a member of the scavenger receptor cysteine-rich (SRCR) superfamily, and is exclusively expressed in monocytes and macrophages. It functions as an acute phase-regulated receptor involved in the clearance and endocytosis of hemoglobin/haptoglobin complexes by macrophages, and may thereby protect tissues from free hemoglobin-mediated oxidative damage. This protein may also function as an innate immune sensor for bacteria and inducer of local inflammation. Alternatively spliced transcript variants encoding different isoforms have been described for this gene. [provided by RefSeq, Aug 2011]
