Background

L-glutamate is the major excitatory neurotransmitter in the central nervous system and activates both ionotropic and metabotropic glutamate receptors. The metabotropic glutamate receptors (mGluRs) are G protein-coupled receptors (GPCRs) that contain 7 membrane-spanning domains and a large extracellular N-terminal region. The group I receptors mGluR1 and mGluR5 activate phospholipase C. By screening a human brain cDNA library with a rat mGluR1-alpha cDNA under low stringency conditions, Minakami et al. (1993) and Minakami et al. (1994) isolated human cDNAs encoding mGluR5. Minakami et al. (1993) identified a longer isoform of mGluR5 (mGluR5B).
