Carbonic anhydrase IX (CA IX) is a transmembrane, hypoxia-induced zinc metalloenzyme that converts carbon dioxide to bicarbonate and protons. This acid-base regulatory function is crucial for both maintaining an alkaline intracellular pH that promotes survival and growth of malignant cells while also amplifying an extracellular acidosis that facilitates their invasion and metastasis (1). Cancer cells that express CA IX often represent the most clinically aggressive components of heterogeneous tumors, making CA IX a key biomarker as well as a major therapeutic target.
